Propylthiouracil inhibits the integration of iodine into the thyroglobulin and with that the thyroxine production.
The thyroid effect results from the inhibition of the intrathyroidal peroxidase-system. Propylthiouracil is well absorbed after oral dose, maximum serum levels will be achieved after 1-2 hours.
Indications/Uses
For the treatment of hyperthyroidism.
Application fields: All forms of thyrotoxicosis, basedow, iodothyroidism, preoperative treatment of basedowfield goiter, completion of radioiodine therapy.
Dosage/Direction for Use
Only a correct and consequent application guarantees a successful treatment and the elimination of side effects. In principle, a difference between initial dose and maintenance dose has to be made.
Usually Propylthiouracil is given every 6-8 hours.
Adults, children and young: Initial dose: 1-2 tablets every 6 hours daily.
Maintenance dose: 1-3 tablets daily.
Children between 6 and 10 years: Initial dose: 1-3 tablets daily.

Use In Pregnancy & Lactation
Pregnancy: During pregnancy, especially during the 10th and 14th week, when the facial production of hormones begins, an especially careful observation is necessary. In the last third, hyperthyreosis often improves spontaneously, that means a reduction of dose may be possible.
Lactation: The concentration in the milk is maximum 1/10 of the mother serum concentration, that is why for nursing patients with hyperthyroid symptoms, Propylthiouracil is accepted as a means of choice.

Gastrointestinal disorders, struma, exophthalmos, leukopenia, thrombocytopenia, agranulocytosis and thyrotoxic crises, after having discontinued the preparation.
